Effect of zinc supplementation

I am taking zinc 8 - 25 mg/day for the last one week along with half tablet/day StresstabsTM.

Compoisition of stresstabs:

Positive effects:

  • Heart rate is around 72-84/min most of the times
  • Blood pressure is also around 120/80 most of the times
  • Reduced gasping for breath
  • Reduced nasal secretions while eating spicy food
Negative effects:
  • Increased lethargy and tiredness

Update on 14 April 2022:

I am taking zinc 15 mg + 400 mcg of methylfolate daily for last 4 days

Positive effects:
1. Less sweating
2. Less anxiety

Zinc is reported to reduce hyperactivity/impulsivity in ADHD patients.

Calcium supplementation increases dopamine

I have taken 1 tablet of neurobion forte multivitamin tablet + 1 tablet folic acid 1 mg daily for 10 days either in the morning after breakfast or after lunch.

Composition of Neurobion Forte Tablet:

Thiamine Hydrochloride (Vit B1) 10 MG

Riboflavin (Vit B2) 10 MG

Pyridoxine Hydrochloride(Vit B6) 3 MG

Cyanocobalamin triturate in gelatin equivalent to cyanocobalamin 15 MCG

Niacinamide (Nicotinamide/Vit B3) 45 MG

Calcium Pantothenate 50 mg

Also, I have taken Calmagic HD tablet for last 5 days along with the above tablets.

Composition of Calmagic HD tablet:
Elemental calcium 500 MG
Elemental magnesium 100 MG
Vitamin D3 1000 IU

I observed the following effects:

1. Decreased tension, decreased anxiety

2. Decreased heart rate (from ~ 90bpm to ~ 82 bpm) and decreased blood pressure (from ~128/86 mmHg ~ 116/78 mmHg)

3. Increased libido and increased erection

4. Slightly increased working memory

5. Increased ability to drive car at faster speeds with better steering control

6. Slightly increased stuttering

All these effects indicate that there is an increased dopaminergic activity. Similar effects were observed during the period 2014-2015 where I took calmagzinc + methylfolate + methylcobalamin.

Effect of folic acid

After stopping centrum energy (contains ginseng extract 40 mg), I have taken folic acid 1 mg daily at night after the meals. I am not taking any other drug/supplements. After stopping centrum energy, I have taken ginger coffee (added commercial ginger paste to coffee) morning and evening for two days. This caused left sided headache suggesting decreased serotonergic neurotransmission. After I stopped ginger intake, that headache disappeared. Taking ginseng for 1 month might have caused increased serotonergic neurotransmission.

Positive effects (probably due to increased doapminergic neurotransmission secondary to disinhibition by serotonergic neurotransmission):

1.  Increased speed of brain processing - I am able to read complex things and understand better and able to remain focused in a long conversation (more than 1 hour). I am able to do things at a faster rate.

2. Decreased post lunch sleepiness

3. Increased libido and strong erections

4. Increased loudness of voice and no hoarseness of voice even after talking for long time.

5. Decreased mucous formation in nose while eating. This increased mucous formation happens whenever I eat lunch despite the food is not spicy and hot. This increased mucous formation may be due to increased cholinergic drive secondary to disinhibition by dopaminergic neurotransmission.

Negative effects:

1. Slight stuttering

2. Slight feelings of stress

3. Increased cold feeling - reduced tolerance to cold climate

Severe headache and ibuprofen

I had severe headache after consumption of becosules Z capsules (just one dose) and applying betnovate N cream for two days. The severe headache may be due to the presence of 100 mg of niacinamide in becosules z capsules. I used to get headache while I was taking niacin before. The headache was right sided indicating that it may be dopamine deficiency-induced. I took 400 mg ibuprofen at midnight. After 40 minutes, the headache disappeared and slept nicely. Earlier, I used to take dicolofenac for such headaches or paracetamol. Diclofenac used to be very effective but due to its enteric coating, it will work after 4 hours only. Paracetamol 650 mg or 1 gm may afford some relief with residual mild headache continuing for next 1-2 days. Ibuprofen seems to be effective. But, I had some stomach discomfort after taking the tablet indicating its ulcerogenic potential.

Natural dopamine antagonists and stuttering

Excessive dopamine in the brain appears to be the causative factor for stuttering in me. As of now, I do not know which part of my brain has excessive dopamine. It may not be really excessive, but may be a relative high level of dopamine compared to other antagonistic neurotransmitters such as serotonin, GABA and acetyl choline in specific parts of the brain which are involved in speech production. Hence, either decreasing dopamine or increasing serotonin, GABA and acetylcholine in those specific parts of the brain may be beneficial in reducing stuttering and achieving fluency. However, as of now, it is not clear exactly in which parts of the brain of stutterers, dopamine is relatively excessive.

In my experimentation with various supplements and drugs, I have observed that the following supplements increase dopamine and worsen stuttering and other issues of speech dis fluency in me. I tried to put them in the order of their effect (descending order). The first supplement causes more stuttering than the second supplement.

Supplements that increase dopamine and increase/worsen stuttering:

1. Omega-3 fatty acids (fish oil) above 100 mg per day
2. Ginkgo biloba
3. Zinc above 6 mg per day
4. Ginseng
5. Vitamin B6 above 6 mg per day
6. Coffee

Supplements that decrease dopamine and reduce stuttering: I tried to put them in the order of their effect (descending order). The first supplement causes maximum reduction of stuttering than the second supplement.

1. Ashwagandha (250 mg extract two times a day; one in morning and one in evening; can feel the effect after 2-3 doses; but it decreases serotonin also; hence, I get negative thoughts)
2. Vitamin C instant release above 250 mg per dose (100 mg taken at night gives better effect next morning)
3. Butter (taken at night gives better effect next morning)

Effects of Ashwagandha

I took ashwagandha tablet (Himalaya) containing 250 mg of Ashwagandha extract in the night after dinner. After few doses, it appeared to have reduced serotonin and dopamine as evidenced by lowered the mood, motivation and initiative. It appeared to have GABA mimicking effect. However, it was stimulatory and did not give me deep sleep. The next day I was not able to focus and concentrate on work. Also, I felt short of breath. While I was climbing stairs and walking uphill, I felt short of breath. However, while speaking, not much of shortness of breath was observed. I could start the sentence easily and maintain the conversation which was slow. An interesting improvement was observed. If one night's dose was skipped, the speech was more fluent and no shortness of breath next day. It appears that GABA inhibits dopamine and serotonin and ashwagandha enhances GABA tone.

Effect of folic acid, methyl folate, zinc and vitamin B6

During the course of several multivitamin tablets and capsules, I observed the effects of few components such as folic acid, methylfolate, zinc and vitamin B6. Actually, these effects are observed when these compounds are taken along with a multivitamin tablet/capsule. Those multivitamin tablet/capsule contain less than daily recommended value of folic acid, zinc and vitamin B6.

When taken alone, zinc 15 mg gives sleepiness and dullness. In contrast, when zinc and vitamin B6 (at least 6 mg) are combined, it gives opposite effect. With this combination, sometimes I don't feel sleepy even around 1.30 am. But If I lie down on bed, I used to get good sleep and complete sleep. Even if it is a 6 hrs sleep, after getting up, even though I still feel the same tiredness and inertia, I don't get the feeling of "not slept enough". Earlier, I used to get "not slept enough....need to sleep for long hours to make up the loss" feeling, if i sleep less than 8 or 9 hours. Because of this feeling, I used to sleep for 8-10 hours during the weekends and 7-9 hours during the weekdays. with zinc and B6 combination, I never worried about sleep debt. Some afternoons, I felt sleepy. But, no urge to lie down and sleep. Earlier, many times I used to go to house and sleep from office. Those times, simply I could not control my sleepiness.

Zinc, when taken alone or in combination gives good muscle flexibility and endurance to sustain 45 minutes of pedaling in cross trainer. However, zinc taken at night disturbs the sleep and the next day I used to feel sleepy. Zinc also heals the skin bruises and scratches fast. My skin around the hand finger nails used to peel off after getting scratched while inserting into my back pant pocket. After taking zinc, simply I don't get this.

Compound
Effect
Remarks
Folic acid
2.5 mg of folic acid gives energy and feelings of pleasure. When listening to music, it gives a very good feeling of intense happiness (just one level below mild euphoria )

Methylfolate
3.75 mg of methylfolate gives mainly enhancement of serotonergic function leading to dullness and sleepiness. On prolonged administration more than 2 weeks, it gives stabilized mood and energy.

Zinc
15 mg of zinc + vitamin C 250 mg
Positive effects
·         good energy and positive mood,
·         reduced day time sleepiness (not completely abolishing but to some extent),
·         feeling of completed sleep
·         Healing of skin scratches and bruises
·         No gasping for breath while exercising, while climbing stairs and while talking

Negative effects
profuse sweating, pronounced stuttering, reduced fluency, non-stutterable pause, slightly reduced motivation, significantly reduced concentration and focus, reduced libido and erections.

7.5 mg – 10 mg of zinc in the form of aminochelate

All the above effects in reduced intensity (dose-dependent reduction) with improved speech fluency and reduced stuttering but still mild non-stutterable pause is there

Without the zinc, I used to feel sleepy and lethargic with any multivitamin combination.

After I observed non-stutterable pause, I stopped taking zinc. The next day, great improvement in energy, mood and almost nil non-stutterable pause was observed.

I googled and found that Zinc can induce Parkinson's like syndrome in animal models by causing death of striatal dopaminergic neurons.

Minocycline Rescues from Zinc-Induced Nigrostriatal Dopaminergic Neurodegeneration: Biochemical and Molecular Interventions.
Vitamin B6 / Pyridoxine HCl
1-5 mg gives moderate energy, focus and concentration. 5-10 mg gives optimum energy, focus and concentration. Above 10 mg gives profuse sweating, sleepiness, reduced loudness of speech and little anxiety due to pronounced stuttering

Effect of bupropion XL

I took bupropion for almost 5 months from January 2014 to May 2014. The comedicines I took were zincovit tablet, mega-3. Initially I felt more energetic and motivated. After 3 months, I did not feel enough motivation and energetic. I used to feel more sleepy. Along with this combination, I took calmagzinc (21st Century brand - calcium, magnesium and zinc). I felt severe headache which lasted for three days. The headache did not go even after taking paracetamol or aspirin. It went away after stopping calmagzinc. So, it appears that magnesium may affect dopamine synthesis and release by inhibiting the glutamatergic neurons.

Also, I read that all antidepressants impair the quality of sperms in males by affecting the DNA and other parameters (motility and count). Bupropion also is reported to affect sperm quality.

Dopamine Biosynthesis

Dopamine biosynthesis
The primary fault in Parkinson's Disease is that, whatever the cause, there is insufficient dopamine. Dopamine is formed in the dopaminergic neurons by the following pathway :
L-tyrosine >>> L-dopa >>> Dopamine
The first step is biosynthesised by the enzyme Tyrosine 3-Monooxygenase [1.14.16.2] (which is more commonly called by its former name tyrosine hydroxylase). The following is the complete reaction :
L-tyrosine + THFA + O2 + Fe2+ >>> L-dopa + DHFA + H2O + Fe2+
So for L-dopa formation, L-tyrosine, THFA (tetrahydrofolic acid), and ferrous iron are essential. The activity of this enzyme is often as low as 25% in Parkinson's Disease, and in severe cases can be as low as 10%. This indicates that one or more of the elements required for the formation of L-dopa are in insufficient quantities.
The second step in the biosynthesis of dopamine is biosynthesised by the enzyme Aromatic L-amino acid decarboxylase [4.1.1.28] (which is more commonly called by its former name dopa decarboxylase). The following is the complete reaction :
L-dopa + pyridoxal phosphate >>> dopamine + pyridoxal phosphate + CO2
So for dopamine biosynthesis from L-dopa, pyridoxal phosphate is essential. The activity of the enzyme rises and falls according to how much pyridoxal phosphate there is. The level of this enzyme in Parkinson's Disease can also be around 25% or even far less.

Coenzymes involved in Dopamine biosynthesis
Besides two enzymes being required for the formation of dopamine from L-tyrosine (L-tyrosine >>> L-dopa >>> Dopamine), three coenzymes are also required. Enzymes are substances that will enable a specific chemical reaction to take place in the body. Coenzymes are substances that assist enzymes. Some enzymes (including those involved in dopamine biosynthesis) will not function without coenzymes.
The three coenzymes involved in the formation of dopamine are : THFA (for L-tyrosine to L-dopa), Pyridoxal phosphate (for L-dopa to dopamine), and NADH (for the formation of THFA and Pyridoxal phosphate). They are made from vitamins via the following means :
Folic acid >>> Dihydrofolic acid >>> Tetrahydrofolic acid
Pyridoxine >>> Pyridoxal >>> Pyridoxal 5-Phosphate (this requires zinc as a cofactor)
Nicotinamide >>> NMN >>> NAD >>> NADH (or NADP) >>> NADPH

PART 3 : CELL DAMAGE

The first step in the formation of dopamine (L-tyrosine > L-dopa > dopamine) is the formation of L-dopa from L-tyrosine.

L-dopa is normally formed in the dopaminergic neurons by the following pathway :

L-tyrosine + THFA + O2 + Fe2+ >>> L-dopa + DHFA + H2O + Fe2+

There are two problems with this reaction not working. Primarily, if somebody does not form L-dopa, they will also lack the dopamine required to prevent Parkinson's Disease from occurring. No L-dopa formation ends up in somebody developing Parkinson's Disease.

However, there is also a cellular symptom of a failure to form L-dopa from L-tyrosine. This is the cell damage that occurs in Parkinson's Disease.

The primary natural means via which cell damage can occur in Parkinson's Disease is due to the reaction from L-tyrosine to L-dopa not taking place. The following is what should happen :

L-tyrosine + THFA + O2 + Fe2+ >>> L-dopa + DHFA + H2O + Fe2+

However, if for example, the THFA in the above reaction is lacking, the following can happen instead :

L-tyrosine + Fe2+ + O2 >>> L-tyrosine + Fe3+ + O-2 (superoxide anion)

As can be seen there is no L-dopa formed in the faulty reaction, and the superoxide anion is formed instead. The superoxide anion is one of the most highly destructive elements in cellsOXIDATIVE DAMAGE.   So you get a double problem : symptoms of Parkinson's Disease and cell damage.

The formation of L-dopa can also fail to take place if L-tyrosine is deficient.

So the simplest means of preventing cell damage from taking place is to ensure that you have those substances required for the formation of L-dopa, which are L-tyrosine, THFA (which is made from the vitamin folic acid using nicotinamide), and ferrous iron.

However, many people have instead tried to solve the problem of cell damage by other means, largely because they did not known the primary cause of the cell damage.

Vitamin C and Vitamin E have been used to try to help to prevent cell damage in Parkinson's Disease. This is because they are claimed to assist in two enzyme reactions in the brain that get rid of the superoxide anion once it has been formed :

Superoxide Dismutase [1.15.1.1] : 2O-2 + 2H+ >>> H2O2 + O2

Catalase [1.11.16] : H2O2 >>> H2O + 1/2 O2

Effect of Vitamin E

I took Vitamin E (250 IU; d-alpha-tocopherol 167.5 mg) in the morning for few days. I experienced severe headache for almost the whole day. However, I observed one strange effect. If I take vitamin E together with a boiled egg, then I did not get any headache.

BIO E 250
Positive effects:

  • Reduced anxiety
  • Able to work continuously with focus and concentration without getting much tiredness and it was helpful for writing articles at one stretch.
Negative effects:
  • Stuttering was mild. However, mild vocal blocks were there during the conversation. The speech was easy to start but after talking for 10-15 minutes, suddenly I was stuck with a vocal block in which I was unable to produce any sound from my throat. I could not even stutter. This vocal block was similar to the one I observed with DL-phenylalanine.
  • Severe headache on right side of my head (indicating dopamine deficiency)
This vocal block might be probably due to the low level of dopamine. There is a research paper supporting this hypothesis.

My view on atypical depression

Since I am suffering with atypical depression, I just analysed the mechanism of neurotransmission probably responsible for the observed symptoms like hypersomnia, hyperphagia and leaden paralysis.

The serotonergic neurotransmission may be normal in some areas of the brain (may be ascending 5-HT neurotransmission) and low in some parts of the brain (may be descending 5-HT neurotransmission which may be from prefrontal cortex and orbitofrontal cortex).

Noradrenergic neurotransmission may be normal (if the person is not having trait anxiety) or may be excessive (if the person is having trait anxiety).

Dopamine neurotransmission may be significantly impaired (both D1 and D2 type).

I feel it is the serotonin to dopamine ratio which is critical in some areas of the brain which is responsible for the observed symptoms.

High serotonin and low dopamine condition (improper ratio) might be leading to hypersomnia. Actually hypersomnia is not a condition of motivated willingness to remain in sleep. Rather it is a condition of unable to motivate oneself from getting up from the bed. So, I feel it is a condition of high serotonin and low dopamine. Because, when the sleep is not refreshing much, nobody will be willing to remain in bed for long period of time. It is the low dopaminergic neurotransmission which might be accountable for this.

The same high serotonin to low dopamine ratio would also be responsible for the hyperphagia. Because, serotonergic drugs induce carbohydrate cravings. So, increased serotonin levels would be leading to carbohydrate cravings thus leading to more food intake and weight gain.

The same high serotonin to low dopamine level would also be responsible for the fatigue, tiredness and heaviness in the legs. I suspect a low dopaminergic neurotransmission in basal ganglia.

Stomach bloating - causes and cures

For the past few weeks, I stopped taking any non vegetarian foods except eggs. During the last week, my stomach bloated (increased in circumference ~ 2-4 inches) made me unable to put on my pants. My belly bulged like a pregnant woman gave me heavy embarassment. Then one day, at lunch and dinner I took two drum stick pieces of chicken. The next day morning, my stomach bloating went away and I was able to put on the same pant without much difficulty. I am suprised to know that my stomach is the root cause of many problems.

I realized that I have stomach bloating when I was 24 years old. I used to have stomach bloating almost throughout the day. But I used to defecate every morning mostly after taking milk tea. No constipation. After defecation, my stomach bloating used to decrease. After taking the breakfast, it used to increase slowly reaching the maximum after lunch, bulging out visibly. In the evenig after taking tea, I used to fart loudly and continuously. The gases in my colon made me to fart loudly. If I didn't fart loudly, I used to feel incompleteness.

From the scientific literature, I understand that, gas formation is responsible for the stomach bloating. However, more than gas formation, it is the hypersensitivity of the intestinal tract to the formed gas matters most. This type of behavior was observed in crohn's patients. So, I checked for symptoms of the Crohn's disease. I never used to have stomach or abdominal pain or constipation. So, I may not have Crohn's disease. I thought I may have inflammatory bowel disease (IBS). So, I checked for IBS symptoms. The Rome III criteria states that abdominal pain or abdominal discomfort is the main symptom and it should be relieved by defecation. I never used to have abdominal pain or discomfort. I used to have only stomach bloating.

Serotonin, dopamine and stomach bloating

I used to feel increased stomach bloating when I used to take serotonergic enhancing formulas like 5-HTP, Magnesium amd chromium. I have to check the effect of fluoxetine on this. When I was taking bupropion, my stomach bloating decreased very much and also farting. No loud farts I did. These observations suggest that my intestinal tract is deficient in dopamine and have relative abundance of serotonin. Improper  serotonin/dopamine ratio exists in my intestinal tract. So, the serotonin produces its maximum effect. Without enough dopamine, my intestinal tract bloats. The fried chicken that I took, might have enhanced the dopamine levels in the intestinal tract thus reducing stomach bloating.

It is also mentioned in the below reference that the abnormal sensitivity of the stomach muscle wall in response to the expansion of intestinal tract may also contribute to stomach bloating.

Suggestions to cure stomach bloating

  • Ensure regular consumption of foods that increase dopamine neurotransmission
  • Do exercises to strengthen the stomach muscles so that their hypersensitivity can be blunted.
  • Consume curd/yoghurt with bacterial cultures such as Bifidobacterium and Lactobacillus (probiotic preparations)

Effect of Bupron (Bupropion SR) - 150 mg

I discontinued fluoxetine and then I took bupropion 150 mg extended release once daily in the morning along with neurogistics multivitamin capsules for 3-4 weeks.

Positive effects:
  • Improved mood - No negative thoughts, No depressed mood
  • Improved energy
  • No procrastination - I could meet the deadline in my job first time in the last 3 years.
  • More focus - can remain in a task for more period of time enabling me to complete the work.
  • More concentration - can think faster and do faster processing
  • No sexual side effects - but no increased libido or strong erections
  • Maximum resolution of anxiety - After 2-3 weeks, I experienced a maximum resolution of anxiety which was haunting me for more than 10 years. No worrying thoughts before driving the car. No worrying thoughts before the class. Sometimes, I used to feel tachycardia. This tachycardia went off after taking few deep breaths.
  • My eye vision improved. I was not taking omega-3 supplements. Even then my vision did not deteriorate. I could see more brightness in the night while driving. Earlier, this lack of brightness was causing me more anxiety while driving in the night.
  • Improved olfactory functions - Taste and smell sensation improved.
Negative effects:
  • More aggressiveness - led to more fights with my wife.
  • No difference in the affection. I was not able to like and love my family (This condition was there before taking medicines. So no improvement in this part.)
  • Music was not enjoyable as like before.
  • Dizziness and syncope
Suggestion:
  • I may need a less dosage of bupropion to reduce the side effects
  • OR I have to include 5-HTP once daily (either morning or in evening) to counteract the probable decrease in serotonin and GABA due to elevated dopamine levels.

    Effects of Mentat

I am taking Mentat tablets (Himalaya drug company) for the past one week (2 tablets in morning and 2 tabs in night). I am also taking multivitamin tablets (Appeton teen grow) and slow release vitamin C tablets (500 mg) + bioflavonoids.

    Positive effects:
    1. Reduced anxiety - may be by increased GABA levels
    2. Reduced stuttering - even though the stuttering is somewhat reduced, no feelings of comfortable and enjoyable fluent speaking. Reduced stuttering was observed for the first 1-2 weeks. After that stuttering was present and sometimes worse.
    3. When I took mutton, it increased my libido and affection.
    4. During 2008, I took mentat for about 1 month. It has increased the libido and affection and love feelings very much especially after I take ground nuts (but did not give rise to strong erections) . So the involvement of omega -6 could be a possible mechanism.
    5. No excessive sleep. I could finish sleeping without much difficulty. I could get up after 7-8 hrs easily. This effect was observed after 1 week of taking mentat. Sleeping was much more difficult with 5-HTP, With 5-HTP, I could not finish my sleeping and my sleeping never seemed to end. I felt like I was unable to motivate myself to get up. So, it indicates serotonin reduces dopamine probably in the mesolimbic region of the brain for motivation.
    6. Increased mobility, increased flexibility of muscles, faster movements without any gait problems, better coordination of muscles while driving were observed after 2 weeks of taking mentat.
    7. I was able to sleep for less time per day, and I was able to motivate myself to do certain things. Example, I had meeting on last week which extended beyond the scheduled time. So I had to rush for the prayer. I could rush for the prayer and I could motivate myself to do such risky operation without any difficulty. I enjoyed such intense stressful rushing. I could drive the car faster and I could reach the prayer place just on time. Even a 2 minute delay would forfeit me from joining the prayers. So it is like "can do" or "cannot do" situation. I could do that. Thats it.
    8. I took less time in getting ready in the morning. usually I take 1 hour 30 minutes (from getting up from bed to start my car; This includes brushing (10 minutes), making and drinking tea (10 minutes) and making breakfast (10 minutes) going to latrine (10-15 minutes), taking bath (20 minutes), eating breakfast (5 minutes), dressing up (15 minutes)  to get ready. After taking mentat, I could get ready by 1 hour by doing all these things faster without having the anxiety of rushing.
    Negative effects:
    1. Muscle weakness - decreased flexibility of muscles (This is minimized by taking vitamin C which may increase noradrenaline levels). But this is not disturbing my driving. This effect was there for first 1-2 weeks. After that, I experienced increased flexibility and mobility. See the positive effects above.
    2. When I started adding DL-phenylalanine 500mg in the morning or in the lunch, it produced headache. Mild headache which lasted for 3 days. This headache did not go away even after taking 5-HTP (50 mg OD). This headache got reduced after taking magnesium + glycine + taurine. I need to explore the mechanism behind this.
    3. After 2 weeks of taking mentat, I got severe headache which is not going even after taking paracetamol 650 mg dose. The headache got reduced after taking magnesium, taurine and glycine and 5-HTP. The headache completely went away after skipping a night's dose of mentat only.
    4. Update on 6 May 2010: I got severe headache yesterday on my right side of head. Due to some purchasing work, I could not take lunch and DL-phenylalanine on day before yesterday. Night also I did not take DL-phenylalanine. Yesterday, I got headache which started around lunch time and worsened in the evening. In the evening, I smoked a cigarette in order to boost serotonin, dopamine and noradrenaline and adrenaline. It gave me a little relief. Then along with dinner, I took DL-phenylalanine 500 mg. Then I applied tiger balm on my side head. Around 12 am headache reduced much and I could sleep better. Today morning, I felt better. So, it looks to me that the headache is probably because of dopamine depletion. Even though I had headache yesterday, my mood was OK and my sociability was also ok. If I get a headache which is aching on left side of my head, then at those times, my mood used to be low. So it could be due to serotonin depletion. yesterday's headache was aching on right side of my head. So, it was different.
Possible mechanisms: (First written on 21 April 2010)

    ObservationPossible mechanism
Less sleep, increased flexibility , less global bradykinesiaIncreased straital and mesolimbic dopamine neurotransmission (Dopamine receptor agonism)
Severe headacheDecrease in serotonin (blockade of 5HT1A autoreceptor)

Possible mechanism (First written on 30 April 2010)

After reading several research papers on anxiety and its animal models, I read about buspirone. The effects of buspirone and mentat seem to be matching perfectly. Since I have not taken buspirone anytime before, I cannot really compare the effects of buspirone and mentat.

See the links below for research articles on buspirone's effects.

Effects of Acute and Chronic Administration of the Serotonin1A Agonist Buspirone on Serotonin Synthesis in the Rat Brain

Alterations of central serotonin and dopamine turnover in rats treated with ipsapirone and other 5-hydroxytryptamine1A agonists with potential anxiolytic properties.

    Update on 12 may 2010:

Mentat consumption along with 5-HTP, DL-phenylalanine and multivitamin capsules produced severe headache which usually lasted for one full day. I had few headache attacks like that in the past week. The headache was mostly on the left side of my head. It was associated with irritation, and psychomotor retardation. So, I decided to discontinue mentat.

According to above source, Mentat has the following pharmacological action.

Mentat regulates cholinergic and GABAergic systems of neurotransmission and thus improves brain functions. It restores activity of muscarine and cholinergic receptors of frontal cortex and thus improves I.Q., memory capacity, concentration, and raises stress resistibility level. Mentat contributes to reduction of tribulin level, an endogenous inhibitor of monoamine oxidase which is increased at anxious conditions of different degree. Preparation stimulates improvement in case of concentration instability and behavioral disturbances. Mentat protects against parkinsonism by raising activity of post-synaptic dopamine receptor. Its sedative and tranquilizing action protects from convulsions and is helpful in insomnia. Mentat improves articulation and corrects speech defects.
